Platelet Activating Factor(PAF) - Induced Peripheral Basophil Histamine Relesability in Asthmatics

Abstract
It has been known that the role of PAF in asthma such as acute bronchoconstriction, submucosal edema, and even bronchial hyperresponsiveness is not mainly direct PAF itself action, but it¢¥s indirect through the other in flammatory cell activation
by
PAF.
The action of PAF in asthma is just similar as that of allergen, and in allergic asthmatic reaction IgE-bearing mast cell and basophils are main primary activating cell.
In this study was observed the PAF(10E-9-10E-5M)-induced peripheral basophil histamine releasability(BHR) in both 5 intrinsic asthmatics and 5 extrinsic asthmatics strongly sensitized to house dust mites, and also checked Dermatophagoides
fariana(D.f.)-induced basophil histamine releasability in extrinsic asthmatics by automatic fluorometric analyser to know that PAF directly induced BHR in asthmatics, so PAF activates basophil such as allergen in extrinsic asthmatics.
@ES The results were as follows:
@EN 1) PAF not induce BHR in both extrinsic and intrinsic asthmatics at any concentration from 10E-2 to 10E-5M.
2. D.f. -induced BHR began at 10E-6 12point and after then it showed dose-response curves in all house dust mite sensitized asthmaics.
3. From these results we confirmed PAF did not induce histamine release from peripheral blood basophil directly, so PAF did no activate basophils in asthmatics.
